Caroline was born in Newtown, Geelong. Her father was an English immigrant and her mother was from Scotland. She had 13 siblings in total. Mockridge won a scholarship to college when she was in sixth grade. She began working as a teacher apprentice in 1893, a profession that she continued with for the next 46 years. She primarily worked in state primary schools. Aside from teaching, she could play various instruments such as the violin, piano and flute.

She was still able to bake scones at age 107. Aged 110, she was described as frail, yet mentally alert. She lived together with her younger sisters until 1982, when she moved to a nursing home. Mockridge never smoked. She never married either.

Mockridge died on 6 November 1987, at the age of 112 years, 330 days. At the time, she was the oldest Australian ever, but she is now the 5th-oldest.
